Maize resistance to insect herbivory is enhanced by silencing expression of genes for jasmonate‐isoleucine degradation using sugarcane mosaic virus

چکیده

Previously, sugarcane mosaic virus (SCMV) was developed as a vector for transient expression of heterologous genes in Zea mays (maize). Here, we show that SCMV can also be applied virus-induced gene silencing (VIGS) endogenous maize genes. Comparison sense and antisense VIGS constructs targeting phytoene desaturase (PDS) showed resulted greater reduction expression. In time course after infection with PDS, lesion mimic 22 (Les22), Iodent japonica 1 (Ij1), efficient observed 2, 3, 4 weeks SCMV. However, at Week 5, Les22 Ij1 no longer significantly reduced compared control plants. The defense signaling molecule jasmonate-isoleucine (JA-Ile) inactivated by 12C-hydroxylation hydrolysis, knockout these leads to herbivore resistance. JA-Ile hydroxylases hydrolases have been investigated Arabidopsis, rice, Nicotiana attenuata. To determine whether the homologs function plant defense, silenced ZmCYP94B1 (predicted hydroxylase) ZmJIH1 hydrolase) SCMV, which elevated two defense-related genes, Maize Proteinase Inhibitor (MPI) Ribosome Inactivating Protein 2 (RIP2). Although increased resistance Spodoptera frugiperda (fall armyworm), Schistocerca americana (American birdwing grasshopper), Rhopalosiphum maidis (corn leaf aphid), there additive effect from both Further work will required more precise functions enzymes regulating defenses.

Sugarcane Mosaic Virus-Based Gene Silencing in Nicotiana benthamiana

Background:Potyvirus-based virus-induced gene silencing (VIGS) is used for knocking down the expression of a target gene in numerous plant species. Sugarcane mosaic virus (SCMV) is a monopartite, positive single strand RNA virus. Objectives:pBINTRA6 vector was modified by inserting a gene segment of SCMV in place of Tobacco rattle virus (TRV) genome part 1 (TRV1 or RNA1)...

Characterization of Sugarcane Mosaic Virus Scmv1 and Scmv2 Resistance Regions by Regional Association Analysis in Maize

Sugarcane Mosaic Virus (SCMV) causes one of the most severe virus diseases in maize worldwide, resulting in reduced grain and forage yield in susceptible cultivars. In this study, two association panels consisting of 94 inbred lines each, from China and the U.S., were characterized for resistance to two isolates: SCMV-Seehausen and SCMV-BJ. The population structure of both association panels wa...

Auxin Binding Protein 1 Reinforces Resistance to Sugarcane Mosaic Virus in Maize.

Dear Editor, Sugarcane mosaic virus (SCMV) causes severe viral diseases in maize worldwide (Fuchs and Gruntzig, 1995), resulting in significant losses in grain and forage yield in susceptible cultivars of maize and related crops. The most promising solution is to cultivate resistant varieties, which contribute to sustainable crop production. Two epistatically interacting major SCMV resistance l...

Enhanced Transgene Expression in Sugarcane by Co-Expression of Virus-Encoded RNA Silencing Suppressors

Post-transcriptional gene silencing is commonly observed in polyploid species and often poses a major limitation to plant improvement via biotechnology. Five plant viral suppressors of RNA silencing were evaluated for their ability to counteract gene silencing and enhance the expression of the Enhanced Yellow Fluorescent Protein (EYFP) or the β-glucuronidase (GUS) reporter gene in sugarcane, a ...
